Abstract: G protein-coupled receptors (GPCRs) regulate a range of physiological processes and thus they are extremely interesting as drug target. Heptares’ proprietary StaR® technology is very valuable for the thermostabilisation of GPCRs in a chosen conformational state. This process generates thermostabilised StaR® protein that can be readily solubilised with detergent, purified and utilised for further applications such as biophysical screening techniques and structure determination via X-ray crystallography. Recently, the usage of StaR® technology at Heptares Therapeutics Ltd. enabled the determination of structures from various GPCRs bound to their respective allosteric modulators.
